Welcome to a bit of theorycrafting, my dear fellow priests. I won't do any number-crunching, I leave this for people who like to do it.
Today we want to talk about the five-second rule, or short the 5SR. Its a thing a good and mana-aware Priest should know about. In essence it comes down to this:
For five seconds after spending mana on a spell, the normal spirit-based mana regeneration will be nulled.
This means that spending mana on a spell, makes you loose mana in two ways:
The spell you have just cast costs mana, which is substracted from your mana pool.
Your mana regeneration will drop for the next five seconds, resulting in less mana gained.
Since loosing mana is a bad thing, you should try to work around the 5SR whenever possible. There are a few tricks to do this:
Cast your Spells in bursts...
Try to squeeze your healing spells together, so that your time between casts may stretch out a bit, allowing more mana-ticks outside the 5SR.
Top off your tank, throw him an additional HoT and finish with a PoM if there is some more damage going round. Then step back and wait until you need to heal again.
Only heal when your heal is needed...
Don't spam heals unless it is absolutely neccessary. Start your spellcasts and interrupt them if the target does not need any healing halfway through your cast. Then start a new cast immediately, even if your target is topped of.
Cast your spells for free whenever possible...
This is the tricky part. A priest, if properly specced, has two abilities at his/her disposal that allow him/her to cast spells for free. Since those clearcasts don't cost mana, they will not trigger the dreary 5SR.
Inner Focus
This ability has a 3 minute cooldown and is found at the 3rd tier in the discipline talent tree. It makes your next spell cost no mana and increases it's critical strike chance by 25%.
Inner Focus
is most effective when stringed together with other clearcasting abilities. This enables you to put out some healing while not straining your mana pool. The longer you are able to evade the 5SR, the better.
Holy Concentration
&
Improved Holy Concentration
This is a passive ability that, when maxed (and you should max it!), will give you a 45% chance to enter a clearcasting state after casting a critical
Flash Heal
,
Binding Heal
or
Greater Heal
.
The clearcasting state lasts for 15 seconds, so you should cast a spell within this timeframe to gain a benefit from it. Do this, even if your target is topped, since the spell can proc of itself and, if lucky, can lead to a string of free heals. As you can see, critical strike rating is becoming a major mana regeneration factor. There is also the talent
Serendipity
that (while providing no 5SR escape) has a nice synergy with your critical strike chance.
If your
Inner Focus
cooldown is up, i suggest you to use it right after you exit the clearcasting stance from
Holy Concentration
. This ensures you more consecutive time outside the 5SR, which is very good.
I'm just trying to clarify some things here. I'm sure pretty many of you out there know this stuff already.

It's there. ;-)
If you look into your armory profile, just check your "Spell" tab for "Mana Regen". On mouseover the tooltip will tell you how much mana you regenerate inside and outside the 5SR.
BTW: The term "while casting" is used by blizzard to describe the time you spend inside the 5SR.

The 5SR is initiated after a cast, that means, although Blizz say "while casting" it isnt infact whilst casting, you can cast and inturupt your cast and still be outside of the 5SR, thus gaining the most out of mana regen but ready to complete the cast if required.

after

Therefore, it is after the cast for non-channeled spells, and at the beginning of the cast for channeled spells.

if not, then the 5SR is normal cause there is another greater rule that says the mana regens with an amount of X every 5 seconds!

MP/5 is separate from the five-second rule, to my knowledge -- it applies at all times, even "while casting".

What other 5 seconds?
The 5-second rule is best thought of as a reduction in mana regen, not a complete nullification. All healing priests should have Meditation to give some MP5 and have MP5 on gear as well.
Your normal idle mana regen is listed as "while not casting" on the character pane. As soon as you spend mana, however, your mana regen will be reduced to the amount listed under "while casting," commonly known as your MP5. You will not resume your full mana regen until you have gone 5 seconds since your last spellcast. Hence, 5 second rule.

Yes, as stated on the spell
Innervate
, it increases your spirit-based mana regeneration by 400% (that means x5), and allows 100% of your spirit based mana regeneration to run, even during the 5SR, effectively removing it for 20 seconds.

A fascinating point.... it has me thinking quite a bit.
In essense, it would seem a far more efficient use of the FSR to cast more channeled spells if your gear is toward mp5 due to the fact that the time that you are channeling you are actually doing something instead of just sitting around. IE, you'll have less downtime due to the 3 seconds that it takes to do the channeling that will count as 3 of your non-generating mp seconds before the mp regen tick starts up again.
Of course, how many channeled priests spells are there to really take advantage of this? I have Starshards and Mindflay that I can think of off the top of my head, which would work well for a SP maybe, but is there a rotation that you could get the channeled frontloading of the FSR to work for you from a healing perspective?
Not sure if I explained that clearly enough or not...
Edit: Maybe saying it like this would be clearer-

The posters are referring to the spell Prayer of Mending and the generic term "Heal over Time".

Yes, you will get the most out of your Spirit if you end your rotation with a channeled (or instant) spell, and start your rotation with a long-casttime spell.

(a GUI mod) gives you a visual representation of the 5SR, you can use this to better understand when you have MP5 WC and when you have full MP5.
