This item only exists within Heroic mode.

As stated above, it is not consumed upon use; however, it is turned in with the quest.

For those interested in lore.. It's probably worth to mention that people are looking for Alleria and Turalyon (Alleria is Sylvanas' older sister who is married to Turalyon a human paladin) The Huntresses could be a link to the Windrunner sisters.

Looted in heroic version of Shadowmoon Burial Grounds
Quest starts and ends from the Inn in your Garrison

Will be removed when you return the quest

It may be worth mentioning that if you go on to kill Ner'zul, and forget to go back to loot the arrow, it seems that you can't get the arrow by running the instance again.
I did this, and the arrow was simply not there, even though I had the incomplete quest in my log. I had to abandon the quest and start again.

Obviously, anecdotal evidence, and not exactly statistically significant, but worth checking out if you are one of those who can't find the arrow.

For those of you who ran the wrong version, you don't need to abandon the quest, just run the Heroic and pick up the spear there instead
